The present invention relates to a DNA library construction method using an adaptor comprising a bar code sequence and to a next-generation nucleotide sequencing method. According to the present invention, in the next-generation nucleotide sequencing, the bar code-introduced adaptor is used, and thus the number of leads is significantly increased compared with a conventional method, thereby sensing even rare mutations which are present at an extremely small frequency.
