The present invention relates to a method for evaluating the activity of a Leucine-rich repeat-containing G-protein coupled receptor 5 (LGR5) activity regulator by using a serum response factor response element (SRF-RE) luciferase and, more particularly to a method for measuring the activity of the luciferase by using the fact that a serum response element (SRE) and the SRF-RE reporter gene expression changes depending on the activity of the LGR5, wherein the activity of the SRF-RE luciferase is not changed by RSPO1, LGR4, and LGR6 but changed only by LGR5. In addition, the LGR5 uses a G12/13-Rho path in order to induce the SRF-RE reporter and, more specifically, controls Extracellular signal-regulated kinase (ERK), focal adhesion kinase (FAK), Nuclear factor kappa B (NFκ B), and c-fos which are downstream control genes of signal delivery.
