INTEGRATION METHODS OF ENRICHING AND DETECTING RARE CELLS FROM BIOLOGICAL BODY FLUIDS SAMPLES

An integration method of enriching and detecting rare cells from biological body fluid samples. The enriching method includes : a) removing plasma protein through centrifugation, b) optionally adding red cell lysate to lyse red cells and remove red cells, c) adding immuno-microspheres or immunoabsorbent and incubating, d) performing special cell separation medium based density gradient centrifugation to separate circulating rare cells from residual red cells and white blood cells binded to the said immuno-microspheres. The method of detecting rare cells enriched includes combining immunohistochemistry based staining with immunofluorescene, or conducting immunohistochemistry based double colored, trichrome or multi-color staining, and utilizing fluorescence or ordinary optics microscope or scanner based on microscope principle to observe and identify. This novel and special enriching and staining method is proved to be low cost, and can quickly, effectively and high specifϊcally enrich rare cells from blood, and quantitatively detect the rare cells.
