The present invention relates to a plant chitosan enhancer composition, comprising Rhizopus oryzae and rare earth minerals, and to a preparation method thereof, which comprises the following steps : liquid-culturing Rhizopus oryzae; decomposing a chitin powder to produce chitosan, by using liquid-cultured Rhizopus oryzae; mixing Rhizopus oryzae culture medium, in which the produced chitosan is contained, with grain and the rare earth minerals, and solid-fermenting a mixture; and drying and pulverizing the solid-fermented fermentation product.
