PROBLEM TO BE SOLVED : To provide methods of detecting a rare mutation in a template DNA in distinction from the mutation caused by the errors in a nucleic acid amplification and nucleic acid sequence analysis, and to provide an apparatus and computer program for performing the method.
SOLUTION : A method comprises the steps of : analyzing the nucleic acid sequence of a library produced by a nucleic acid amplification reaction using a sample containing a template DNA of 1000 copies or less; calculating the mutant ratio in a base at a given position from the analyzed results; and comparing the calculated mutant rate with a given cutoff value to detect a rare mutation.
