PURPOSE : A method for preparing a single cell-derived genome library of rare microorganism or novel microorganism based on a technique for removing dominant species is provided to enable to confirm origin of a gene.CONSTITUTION : A method for preparing a single cell-derived genome library of rare microorganism or novel microorganism comprises : a step of performing microorganism group community analysis of collected sample; a step of labeling dominant or known microorganism in the community analysis; a step of removing labeled microorganism by primary FACS(Fluorescence Associated Cell Sorter) method; a step of labeling remaining microorganism group and isolating total microorganism group into single cells by secondary FACS method; a step of amplifying the genome form the single cells by MDA(multiple displacement amplification); and a step of preparing a genomic library.COPYRIGHT KIPO 2011 |
