PROBLEM TO BE SOLVED : To obtain new rare-cutting endonucleases which have new sequence specificity about recognition and cleavage.
SOLUTION : There are provided hybrid and/or single-chain rare-cutting endonucleases, called meganucleases, which recognize and cleave a specific nucleotide sequence; polynucleotide sequences encoding the rare-cutting endonucleases; a vector comprising one of the polynucleotide sequences; a cell or non-human animal comprising one of the polynucleotide sequences or the rare-cutting endonucleases; and a method for producing one of the rare-cutting endonucleases and any use of the products and methods. More specifically, any use of such rare-cutting endonuclease for genetic engineering and gene therapy is contemplated.
