A detection method for enhancing the rare earth complex fluorescence based on nucleic acid hybridization, comprising : performing intermediate modification on a first nucleic acid strand, adding a rare earth complex to obtain a coupling product of the first nucleic acid strand and the rare earth complex, and then adding a second nucleic acid strand complementary to the first nucleic acid strand to achieve the effect of enhancing fluorescence. The method has the advantages of simple reaction operations, green mild conditions, short reaction time, etc. The fluorescence of a coupling product of nucleic acid and the rare earth complex can be enhanced up to more than 20 times in the presence of nucleic acid complementary strands. The method can be used for detecting various nucleic acids, proteins and other small biomolecules at high sensitivity in a physiological condition.
