PROBLEM TO BE SOLVED : To provide rare-cutting endonucleases, also called custom-made meganucleases, which recognize and cleave a specific nucleotide sequence. SOLUTION : There are provided : meganuclease variants derived from an initial meganuclease presenting a recognition and cleavage site different from the site of the initial one; polynucleotide encoding the meganuclease variants; recombinant vector cell, animal or plant comprising the polynucleotide sequences; and uses thereof for in vivo or in vitro genetic engineering and genomic engineering.
