The present invention relates to a method for producing a DNA library using an adapter comprising a barcode sequence and a next generation sequencing method. According to the present invention, the next generation nucleotide sequencing uses the adapter into which the bar code sequence is introduced, when being compared to a conventional method, even a rare variation existing in extremely small frequency can be detected by significantly increasing the number of reads.
